Title: PROXIMATE ANALYSIS OF BANANA STALK AND CORN COBS
ABSTRACT
The proximate analysis is the approximate estimation of different nutrients of food which includes moisture, ash, lipid, protein, and carbohydrate contents. The present study was designed to find out close relationship between the different components and objectives of study of the both samples. The moisture, dry, ash, oil, crude fibre, protein, carbohydrate, starch and vitamin C content will be estimated by analytical processes Association of official Analytical Chemists (A.O.A.C). The data obtained will be analyzed by statistical methods like complete randomized design (C.R.D) and F test. The analytical procedures will be carried in the Department of Biochemistry labs of Pir Mehr Ali Shah Arid Agriculture University Rawalpindi (Pakistan). The importance and relationship of both samples will be analyzed by comparing the components of banana stock and corn cobs. All the waste samples considered as a great values even greater than some well-known biomass-fuels fall within the limits for the production of steam in electricity generation. These wastes were also used in production of different industrial enzymes which is a matter of great importance for national interest. These waste materials are also used as fertilizer for different crops.
this is my research abstract during my M.Sc.
now i am in M.Phi in biochemistry from PMAS UUAR and my reseach topic is
Hyperproducion, isolation, purification and characterization og glucomylase by various fungi
Abstract
Glucoamylases are used mainly in the production glucose syrup, high fructose corn syrup, and in whole grain and starch hydrolysis for alcohol production. This paper reviews the status of glucoamylases with respect to microbial sources, biochemical and physical properties. Methods used to assay glucoamylase activity are also compared, with reference being made to the specificity of spectrophotometric methods in detecting end products of the enzyme action. Commercial glucoamylases and development of immobilization techniques are discussed. Also, structural analysis of glucoamylase and the main amino acids involved in catalysis and starch binding are emphasized. The cloning of the glucoamylase gene in Saccharomyces cerevisiae for the brewing of low calorie beer is presented. This review highlights the use of glucoamylase in the food industry. Several types of enzymes are involved in the degradation of starch. Glucoamylase, which attacks both a (1 – 4) and a (1- 6) linkages of starch are the most important and accordingly have received the most attention. The catalytic domain of glucoamylases G1 and G2 from Aspergillus niger is produced in vitro in high yield by limited proteolysis using either subtilisin Novo or subtilisin Carlsberg. Glucoamylase will be purified from Neurospora sitophila, cultured on wheat bran, through solid-state fermentation. Maximum enzyme activity obtained will be in IU/ml/min, under pre-optimized media conditions. At 10% (NH4)2SO4 saturation maximum activity will be obtained in filtrate i.e. after gel filtration (Sephadex G-200) protein contents will be calculated. Molecular weight of enzyme will be determined with SDS-PAGE. Optimum temperature and pH will be 50 °C and 5, respectively. Magnesium (Mg), Zinc (Zn), Manganese (Mn), Calcium (Ca) activity will be determined. Different kinds of chemicals will be used to create mutation in particular strain. The mutant produced an appreciable yield of glucoamylase in the culture medium. The enzyme will be purified and will be shown to be homogeneous to desire one. It will hydrolysed successive glucose residues from the non-reducing end of starch molecules. The purified enzyme will have an optimum pH will be seen. It will be determined that at which temperature, it is highly active and its optimal activity. At the end, molecular characterization of the genome of the strain will be determined which shows maximum activity.
