The molecular analysis of the human nucleotide-receptor P2Y11 serves to develop selective ligands. This is important for the medical application of this receptor in various diseases.
Nucleotides are important for cell communication, –development und –survival as signaling molecules. Nucleotides act via P2Y- Receptors (P2Y-R), a subfamily of 7-transmembrane domain receptors. Of the 8 human P2Y-R, the P2Y11-R, which until now is very little investigated, has a distribution, which makes it a candidate for cardial diseases and immune processes.
The amino acid residues in the protein sequence of the P2Y11-R essential for ligand binding are studied by defined Mutagenesis und Molecular Modelling. The physiologic significance of the P2Y11-R should be clarified in immune cells and astrocytes. Also the influence of the Heterodimer formation, which we have discovered for the P2Y11-R with the P2Y1-R, should be investigated.
The Project has molecular biologic and cell biologic spectrum of methods.
