Job Description: Mitochondria are essential for the survival of mammalian cells. These organelles are involved in numerous critical functions including oxidative phosphorylation and the transduction of energy. It is, therefore, not surprising that mitochondrial dysfunction is associated with metabolic disorders, neurodegeneration and with the ageing process. To improve our understanding of how mitochondrial dysfunction contributes to these processes it is crucial that we increase our knowledge of the basic molecular mechanisms that dictate healthy mitochondrial gene expression.

This project aims to investigate :-
• mitochondrial protein synthesis
• how this is regulated
• whether, as an individual ages, there is a change in this regulation
• whether this regulation has tissue specificity

The new technique of ribosome profiling allows quantitative analysis of translation of each of the mt-transcripts. Using this approach will also monitor stalling of mitoribosomes and how this may be affected by known pathogenic mt-DNA mutations. Further, this technique will be used to characterise changes to the mt-gene expression profile as a function of age. Investigations would initially be performed using cell lines derived from young versus older individuals, and if time permits, comparing brain to skeletal muscle to identify if there is a tissue specific pattern to mt-gene expression.

Value of the Award and Eligibility
This award is available to students who meet the research councils’ eligibility criteria for a full award, which covers tuition fees at the UK/EU rate and an annual stipend of £13,290 (2009/10).

Person Specification
You must have, or expect to achieve, an upper-second-class Honours degree, or equivalent, in a relevant subject.
