International PhD Position: Analysis of the Role of the Eukaryotic SRP Receptor in Protein Translocation, Germany

Job Description: The endoplasmic reticulum is a major site of protein synthesis and represents the entry point to the secretory pathway. Since about 30% of all proteins in a eukaryotic cell, including hormones, receptors, channels and pumps, commence their biogenesis at the ER, this is a fundamental cellular process. Disruptions in the early stages of synthesis of these classes of substrates are frequently associated with disease. Understanding, the detailed dynamics of protein targeting and translocation in healthy cells is therefore critical if we are ultimately to intervene to potentially ameliorate such diseases. The thesis project aims at a better understanding of the role of the eukaryotic SRP receptor at the interface between protein targeting by the SRP system and membrane insertion/translocation. In particular the project addresses the interaction of the SRP receptor with the translocation channel and its influence on translocon architecture.
Methods that will be used:
Protein engineering, expression, purification, spectroscopy, membrane insertion assays, protein crystallization and X-ray crystallography

Profile of candidate‘s qualification:
Background in protein biochemistry, molecular biology and/or biophysics, structural biology; interest in protein targeting mechanisms and in interdisciplinary research.
