PhD Student Position in Immunology, Ulm, Germany
For the following DFG funded projcet we search for a PhD candidate with experience in cellular immunology. The funding period will last 3 years.
In the well-controlled transfer colitis model, we intend to identify co stimuli that promote monoclonal, colitogenic CD4 T cell responses locally induced by mucosal DC presenting the cognate antigen delivered orally. The phenotype of intestinal DC (subsets) from SPF and GF mice and their capacity to present orally delivered ovalbumin (OVA) to CD4 T cell populations will be tested. Continuous OVA feeding of SPF RAG-/- mice reconstituted with naïve OT-II RAG-/- CD4 T cells induces a progressive, lethal IBD. The IBD heals when OVA feeding is discontinued, and resistance to reactivation of the disease by oral rechallenge with the antigen develops. We will search for phenotypic changes in DC as well as (colitogenic versus tolerogenic) OT-II RAG-/- CD4 T cells (from the cLP, MLN, spleen and liver) that correlate with IBD development, healing of IBD or resistance to IBD induction. Attempts will be made to define TLR-mediated, colitogenic co stimuli by delivering OVA orally through !
mutant E.coli strains to SPF RAG-/- mice reconstituted with naïve OT-II RAG-/- CD4 T cells. Candidates will be verified by testing if oral codelivery of TLR agonists with OVA can induce IBD in GF RAG-/- mice reconstituted with naïve OT-II RAG-/- CD4 T cells.
