Job Description: Cobalamins (Cbls, vitamin B12 derivatives) are essential cobalt-containing cofactors for two enzyme reactions in humans. MeCbl-dependent methionine synthase transfers a methyl group from methyltetrahydrofolate to homocysteine to generate tetrahydrofolate and methionine. AdoCbl-dependent L-methylmalonyl-CoA mutase converts L-methylmalonyl-CoA to succinyl-CoA. Humans obtain Cbl (1-6 microgram) from animal products, however as many as 10-40% of the elderly are B12 deficient.
Oxidative stress is a condition associated with overproduction of Reactive Oxygen Species (ROS) such as free radicals and peroxides, which attack and damage cellular components including proteins, DNA and lipids. Oxidative stress has been implicated in a wide range of diseases associated with aging, including cardiovascular and neurological diseases. A new, emerging field concerns the non-enzymatic role of Cbl in attenuating oxidative stress. This project is focused on determine the rates and mechanisms of the reactions of Cbls with a range of ROS. A wide range of instrumentation and techniques will be used, including stopped-flow, UV-vis and nmr spectroscopy, HPLC, mass spectrometry and air-free techniques. We will also determine the ability of Cbl to attenuate elevated levels of ROS in mammalian cells and to protect against ROS-associated cell death. Students may choose if they also wish to become involved in the cell studies.
