Our goal is to understand the mechanics, energy transduction and regulation of functional molecular motor complexes in living cells. In particular, we are interested in the molecular machines that transport DNA and type IV pili through bacterial cell envelopes. Molecular motors are nanomachines that move objects by repetitive motion on polymer filaments. To this end, they transform chemical energy into motion. They are involved in the transport of cellular organelles, cell motility, DNA processing and transport of molecules through membrane pores. The length scale of an elementary motor step is set by the size of the motor proteins, i.e. nanometers. Molecular motors generate forces ranging from piconewtons to 100 piconewton. To characterize kinetics and force generation of molecular motors at the single molecule level, we have to measure nanometer-lengths and piconewton forces. Currently we are recruiting a physicist for nanomanipulation experiments using optical traps. The aim will be to characterize the molecular mechanism of force generation by bacterial motors called pili at the single molecule level. Pili are among the most important and ubiquitous bacterial virulence factors and we have shown that they generate remarkably high molecular force in the range of 100pN. To elucidate the mechanism and control of force generation, physicists, biologists and biotechnologists collaborate in our research group.
