PhD Position: Role of the membrane-bound bile acid receptor TGR5 in the liver

The project A10 entitled „Role of the membrane-bound bile acid receptor TGR5 in the liver” is looking for a highly motivated PhD student. The projects investigates the function of the novel, G-protein coupled bile acid receptor TGR5 in the liver. Bile acids are not only essential for dietary lipid absorption and cholesterol excretion, but are signalling molecules with endocrine functions. Bile acids can activate intracellular signalling pathways and modulate gene transcription through nuclear bile acid receptors. TGR5 is a G-protein coupled, plasma membrane bound bile acid receptor, which is expressed in many human and rodent tissues, including different non- parenchymal cells of the liver (Keitel et al. Hepatology 2007, Keitel et al., BBRC 2008). Aim of the project is to elucidate the expression, localization and function of the membrane bound bile acid receptor in the liver. TGR5 activated signalling pathways will be investigated in isolated liver cells and transfected cell lines. Using TGR5 knockout mice the function of the receptor will be studied in different models of cholestatic liver disease.

Methods: Standard techniques of molecular cell biology (e.g. cloning of cDNA, insertion of mutations using site-directed mutagenesis, quantitative realtime PCR), cell culture techniques and transfection, immunofluorescence staining and confocal laser scanning microscopy, live cell imaging FRET and TIRF microscopy to study protein-protein interactions and trafficking of the receptor. Isolation of different cell types from rat and mouse liver and animal experiments.

We offer close supervision in an interdisciplinary team and stimulating environment as well as participation in our “Graduiertenkolleg” with seminars and practical courses.
