Project:
Glomerular proteinuria, normally prevented by the permselective glomerular filter in the kidney, is an indicator for the development of end-stage renal failure and cardiovascular complications. The aim of the project is to decipher mechanisms leading to the loss of protein through the renal glomerulus.
We will analyze a unique data set of already established glomerular gene expression profiles from patients with the main acquired proteinuric glomerulopathies. A recently generated library of genes predominantly expressed in the glomerulus will be used to identify molecular mechanisms particularly relevant in this renal compartment.
Using these data sets regulatory pathways linked to specific or all proteinuric diseases will be analyzed using bioinformatic characterization. This will lead to confirmatory studies in vitro on cell lines of all relevant glomerular cell types. Pathway interference in these cells is planned using siRNA as well as transient or stable transfection. Computer-predicted promoter characteristics will be evaluated by promoter-reporter construct analysis and could be confirmed by DNase footprinting and chromatin immune precipitation. Further evaluation by immunohistology can be performed on an archival biobank.
In parallel, marker transcripts for clinical outcome and response to therapy will be identified by correlation of gene expression profiles with already available clinical follow-up data.

Profile:
We are looking for a friendly and highly motivated new member in our team. A degree in Biology, Biochemistry or Medicine would be ideal.
The work would be partially computer-assisted (array analysis, pathway identification, promoter analysis) and wet bench (glomerular cell studies, siRNA, transfection, if interested promoter interference and/or immunohistology). No animal studies planned for this period of the study.
