PhD Scholarship in Genetic Systems to Characterize the Host Impact of Commensal Intestinal Anaerobic Bacteria,UK

Job Description:The human large intestine harbours a large and diverse resident bacterial population that is important in maintaining health. One function of the gut microbiota is the fermentation of dietary foodstuffs releasing short chain fatty acids that can provide energy to the colonic epithelial cells, but which also interact directly with specific receptors on the epithelial surface. Ongoing genome sequencing projects are providing a wealth of new genome information about both single bacterial strains and bacterial communities. However functional studies on cultured bacteria are essential to confirm gene function and bacterial interactions. Currently there are limited techniques available to perform genetic manipulations on the vast majority of commensal anaerobic bacteria.

In this PhD studentship proposal we intend to establish genetic manipulation techniques to enable us to knockout specific bacterial genes, confirming their function in vitro and in vivo, and comparing the effects of the wild-type and mutant bacteria on host cells and interactions with other bacteria.

The project will draw upon the internationally recognized expertise of the Rowett Institute (Aberdeen, Scotland) and the Alimentary Pharmabiotic Centre (Cork, Ireland), and the appointed student will spend approximately half of the PhD research period at each institution, under the supervision of Dr. Karen Scott and Dr. Paul O’Toole, respectively. The appointed student can expect to learn a diverse range of techniques in prokaryotic genetics including PCR, cloning, sequence analysis, qRT-PCR and microarray analysis, and gene knock-out technology. A range of microbiological and cell biological analyses may also be employed.
