Title: Understanding the structure and function of the ubiquitylation machinery
Project Description: One PhD scholarship is available for a student interested in studying the biochemical properties of ubiquitin E3-ligases in A/Prof. Catherine Day’s laboratory in the Biochemistry Department, University of Otago, New Zealand.

My laboratory has focused on the analysis of protein complexes that have an important role in regulating apoptotic signaling pathways (see http://biochem.otago.ac.nz/staff/day/cday.html). Most proteins are modified by the addition of ubiquitin, yet much remains unknown about the structure and function of the proteins that promote attachment of ubiquitin. We are interested in understanding the structural properties of E3-ligases and how differences in structure underpin differences in activity. Eventually we hope that greater understanding will allow proteins to be manipulated for treatment of disease. The proteins of interest will be produced in E.coli and the aim of our research is to understand their function at the molecular level and to undertake a biochemical analysis of activity. The position offers opportunities to start a career in an area of increasing importance in biological research.

The University of Otago is the top ranked universities in New Zealand. The Biochemistry Department is well equipped for protein based research and houses state-of-the-art research platforms for mass spectrometry, crystallography and protein analysis (ITC, CD). My laboratory is interactive and well resourced.

Suitable candidates should hold a Masters Degree (or equivalent) in Molecular Biology or Biochemistry with a minimum GPA of 8, or equivalent. Successful applicants will be excellent team players. An understanding of the importance of protein chemistry (purification, analysis including crystallographic approaches) cell signaling pathways and molecular biology (cloning, PCR, mutagenesis), as well as basic computer skills would be an asset and previous experience in these areas, although not essential, would be an advantage.

Starting date is flexible but early in 2010 is preferable.
