The research group of Dr. Gert Jansen is situated in the Department of Cell Biology, headed by Prof. Dr. Frank Grosveld, of the Erasmus MC, Rotterdam. The Department has a modern infrastructure that is shared with a number of other Departments in the faculty (Genetics, Biochemistry, Reproduction and Development, Forensic Molecular Biology, Bio-informatics and Clinical Genetics). The Department has an international PhD program (approximately 30 students from many different countries).
The regulation of intraflagellar transport in the cilia of mammalian kidney cells. In the kidney, cilia monitor fluid flow and regulate cell proliferation and differentiation. Disruption of cilia function
can cause polycystic kidney disease. The development and function of cilia require transport structural components and signalling molecules along the cilia, mediated by intraflagellar transport (IFT). Recent results
suggest that IFT is regulated, allowing modulation of cilia length and organization of signalling pathways in cilia. We recently identified a conserved MAP kinase, DYF-5, which is involved in the regulation of IFT in the nematode
C. elegans (Burghoorn et al. 2007, PNAS 104, 7157). Mammals have three DYF-5 homologues. We expect that at least one of these has a similar function in the regulation of IFT. The PhD student will study the regulation of IFT in mammalian kidney cells using life imaging of IFT and knockdown and overexpression of the mammalian homologues of genes that play a role in this process in C. elegans.
Requirements:
We are looking for an enthusiastic and motivated young scientist who has recently completed his/her MSc (doctoraal) in Biology or a similar discipline. The candidate should preferably have some knowledge of and experience with imaging and molecular biological techniques. The department of Cell Biology is internationally
orientated, so English language skills are essential.
