Job Derscription: The department of Biomedical Engineering of the Eindhoven University of Technology integrates engineering and biomedical sciences to investigate one of the most complex and fascinating “systems” known to date: the human body. Within this department, a prominent research field of the Soft Tissue Biomechanics and Tissue engineering group is the engineering of functional, living cardiovascular tissues. In particular the engineering of living heart valves is explored.

Around 24.000 people die annually of an acute myocardial infarction or stroke in the Netherlands. At present approximately 1 million individuals have survived a stroke or myocardial infarction, many of which suffer from serious morbidity. Acute cardiovascular events, such as an acute myocardial infarction, and cerebrovascular events, such as stroke, are caused by rupture of a vulnerable atherosclerotic plaque. A plaque at risk consists of a necrotic core covered by a thin fibrous cap, the presence of inflammatory cells and/or blood cells in the plaque. The inflammation will weaken the cap, which may cause rupture. Rupture of the fibrous cap brings the necrotic core in contact with the blood which cause the formation of a thrombus that may occlude the vessel. Atherosclerosis is a silently progressing disease as for many people an acute cardiovascular or cerebrovascular event is the first symptom of this disease.

Tasks

A PhD position is available in this project to investigate quantitatively the relation between the biological composition of the plaque to rupture characteristics of the collagen network in the fibrous cap. To explore this, we use of a dedicated experimental technique; a recently developed indentation device mounted on top of a confocal laser scanning microscope (CLSM) that enables local measurement of the anisotropic mechanical and failure properties of the cap at the micron scale. In combination with a collagen specific fluorescent probe the CLSM can be used to visualize deformation of the collagen fibers real-time during the experiment, and serves as necessary input to the inverse computational method to identify the mechanical properties.
