A PhD student position (TV-L E13, 50%) is available at the Institute for Neurophysiology (Group Dr. Kuzmenkin), initially for 1 year, then to be prolonged after positive evaluation.

Project “Functional characterization of induced pluripotent stem (iPS) cells and their differentiated derivatives”

Description
Embryonic stem cells (ESC), due to their pluripotency, can be differentiated to various tissues, thus representing a powerful tool for cell-regenerative therapy. However, sources of ESC are sparse and ethically controversial. Another problem is a lacking histocompatibility leading to a rejection of the transplanted tissue. Recently, terminally differentiated somatic cells, fibroblasts, were reprogrammed to an ESC-like pluripotent state by ectopic expression of defined factors. This approach can revolutionize the modern medicin if any “customized”, patient-specific tissue can be generated by such reprogramming of somatic cells of a patient and a following differentiation. However, it is unclear whether induced pluripotent stem cells (iPS) can give origin to a functional tissue, particularly to cardiac muscle and whether the generated tissue has the same physiological parameters as the normal one. As expression of voltage-gated ion channels can be used as a marker of cardiac d!
ifferentiation, we plan to determine expression patterns and functionality of voltage-gated ion channels on every stage of the differentiation process and compare them to ESC-derived as well as to fetal cardiomyocytes. Additionally, iPS cell-derived cardiomyocytes will be tested for unimpaired humoral regulation and sensitivity to channel blockers, as well as for mature action potentials.

Experiments will be done using electrophysiological (patch-clamp), as well as molecular biology (e.g. PCR) techniques. Further, cell culture work and cardiac differentiation will be routinely performed.
The successful applicant should hold an equivalent of a Master’s degree or Diploma in Natural Sciences. Very good communication skills in English and good MS-Office skills are prerequisites. We expect high motivation and creativity. Knowledge in molecular biology techniques and cell culture work would be an advantage.
