December 8, 2008A postdoctoral position is available in the development biology research program at the Lund Stem Cell Center. The research projects focus on pancreas development. In vivo (mouse models) and in vitro techniques will be used to investigate the mechanisms underlying endocrine cell expansion, differentiation, and maintenance. The Stem Cell Center offers strong research profiles in basic (developmental biology, stem cell biology, and molecular biology) and translational research. The pancreas developmental biology program consists of two dynamic groups with joint research interests in cell signaling and transcriptional regulation during pancreas development. We are seeking highly motivated researchers who have recently completed their doctoral programs. Experience in molecular biology or developmental biology is required. However, potential for scientific excellence will be the primary selection criterion.

Production, purification and characterization of chitinase from Streptomyces sp. PTK19 and its biocontrol efficacy against Fusarium wilt of Lycopersicon esculentum
Abstract of Ph.D., thesis:
Isolation and screening of chitinase producing Streptomyces sp. from soil and optimization of cultural conditions at different substrate concentrations, pH, temperature, nutrients namely carbon, nitrogen and amino acid supplementation under shaking condition. Attempts were also made for the production of chitinase in laboratory scale stirred bioreactor. The chitinase obtained from the liquid culture broth of Streptomyces sp. PTK19 was subjected for purification. The enzyme was purified through Ion exchange chromatography and gel filtration chromatography techniques. The molecular mass of purified chitinase was determined at 31.1 kDa using SDS-PAGE by comparing the mobilities of standard protein markers run concurrently. The properties namely pH, temperature optima, substrate specificity, stability and metal ions and chemical reagents inhibitors of purified chitinase were recorded. The Fusarium oxysporum PTK2 strain was used for the pot experiments. Dual plate method was conducted, the pot study experiments proved the biocontrol efficacy of Streptomyces sp. PTK19 against Fusarium wilt of Lycopersicon esculentum. The incidence of disease was measured on the basis of wilt symptoms. The Induced Systemic Resistance (ISR) components of the defense enzyme content were analyzed like peroxidase, polyphenoloxidase and chitinase from the leaf samples of Lycopersicon esculentum at 15, 20 and 25 days of old plants after transplantation and the experiments were conducted for a period of 90 days. The biocontrol efficacy of Streptomyces sp. PTK19 was proved.
