Titile  : Protein biomarkers for human radiation exposure 

Description  : The aim of this project is the development of novel approaches for assessing radiation exposure in samples of biological material from exposed individuals. The hypothesis is that semi-quantitative analysis of a combination of protein biomarkers in blood samples can determine radiation exposure levels with sufficient accuracy and speed to facilitate field triage of large numbers of potentially exposed individuals. Using an antibody for the phosphorylated histone gamma-H2AX which indicates the presence of DNA damage (Rogakou et al, J Cell Biol, 1999), we have previously quantified radiation-induced DNA damage in cells exposed to X-ray doses as low as 1 mGy (Rothkamm & Lobrich, PNAS, 2003). This methodology has recently been applied to patients undergoing computer tomography scans to demonstrate its usefulness as sensitive and quantitative biomarker of human low-level radiation exposure (Rothkamm et al., Radiology, 2007). Furthermore, we have now developed a microscope system for automated cell finding, image acquisition and quantitative analysis of fluorescent markers to increase the throughput (Barber et al., Proc SPIE, 2007). In addition to gamma-H2AX, a number of other proteins and protein modifications will be evaluated as candidate quantitative markers for radiation exposure. The rationale is that a combination of several biomarkers with different kinetic and dynamic properties in response to radiation would provide a more robust result than analysis of one single marker. This project is highly interdisciplinary, encompassing radiation biology, biochemistry, haematology and cytogenetics and utilising sophisticated optical analysis techniques. Collaborative groups involve the Centre for Cancer Research & Cell Biology, Queen’s University Belfast and the Gray Cancer Institute, University of Oxford.
