Quantitative mass spectrometry of oligomeric biomolecules
Description : Analytes often form a group of homologues, regio and/or stereoisomers. Determination of group parameters does not take into account that biological activities – e.g. toxicity, mutagenic activity, antioxidant activity – are strongly related to the individual constitution and spatial orientation of a molecule and can thus differ significantly from related compounds. Mass spectrometry is a powerful tool to gain structural information on tiny amounts of complex mixtures, but cannot simply be used for quantification. Discrimination in mass spectrometry depends on sample preparation, ionisation conditions, ion transfer into the mass analyzer and type of analyzer. By means of appropriate labelling strategies, structural differences influencing ionisation yield can be overcome. Quantitative analysis of complex mixtures of, for example, oligosaccharides and polyphenols will be possible with appropriate instrumentation. Composition behind isobaric signals will be evaluated by tandem mass spectrometry. Diastereoisomers can be differentiated by their metal-coordinating properties, e.g. of diols or other chelating structural elements. Prof. Dr. Petra Mischnick You are required to hold an excellent university degree (master’s or equivalent degree) in physics, chemistry, electrical or mechanical engineering, mathematics, information technology or related fields. The entire program is run in English, so fluency in both written and spoken English is required. Skills in German are welcome. You may be a member of and receive a scholarship from the IGSM for up to 3 years. In your first semester, you will be accepted on a probationary basis.
