Regulation of Sphingosine Kinases/Calcium Signalling by Sphingosine-1-phosphate

The lipid mediator, sphingosine-1-phosphate (S1P), regulates cell proliferation and migration, and is involved in processes such as angiogenesis, vascular contractility and permeability, lymphocyte trafficking, and inflammation. S1P is formed by sphingosine kinases and degraded by phosphatases and S1P lyase. The thesis projects are:
1. Regulation of activity and subcellular localization of sphingosine kinases. Aim is also to correlate the subcellular localization  of S1P formation with a specific cell response, e.g., activation of S1P receptors.
2. Regulation of Ca2+ homeostasis by sphingosine kinases and S1P lyase. Knockout of these enzymes leads to perturbations of the cellular Ca2+ homeostasis. The underlying mechanisms and functional consequences have to be further analysed.

Methoden:
- Cell culture including primary cell culture
- Confocal microscopy / live cell imaging including Ca2+ imaging
- Diverse biochemical methods such as Western blot, sphingosine kinase assays etc.
