Swansea University’s School of Medicine- BBSRC PhD Scholarship, UK
Job Description: Swansea University’s School of Medicine is offering a 4 year PhD studentship funded by BBSRC DTG CASE for a project focussing on “Analysis of the role of ARF6 in GLP1 receptor-mediated insulin secretion, islet cells proliferation and the receptor trafficking” for October 2010 entry.
Glucagon related peptide (GLP)1 hormone binding to its receptor, GLP1 receptor (GLP1R; a Gas coupled GPCR), on the surface of pancreatic b cells results in glucose-dependent insulin secretion and promotion of b cell proliferation. These functions make the GLP1R as an important drug target for the treatment of type 2 diabetes. The internalisation and intracellular trafficking of GPCRs play a critical role in regulating their function through signal desensitisation, receptor recycling, and down-regulation. ARF6 small GTPase and its regulators play important roles in GPCRs internalisation and signalling. ARF6 also regulates insulin secretion. However it is not known whether ARF6 regulate GLP1R receptor internalisation and trafficking, and GLP1R-mediated insulin secretion and islet cell proliferation.
The studentship project will make use of human cell lines and tissues that express GLP1R endogenously or exogenously to study the role of ARF6 in GLP1R internalisation and agonist-induced insulin secretion and islet cell proliferation. The project will also utilise cell biological (immunofluorescence and live cell imaging), biochemical (ELISA, surface biotinylation and immunoprecipitation) and molecular biological (siRNA and RT-PCR) methods. The student joining our laboratory in the Diabetes research group for this project will be together with several PhD students and post docs, and will work in the state of the art laboratories in the new Institute of Life Science.
